When freighted with oxygen it becomes oxyhaemoglobin and is red in color; when freed from its oxygen it takes a purple hue; hence the widely different appearance of arterial and venous blood, which so puzzled the early physiologists.
That the oxygen forms a definite compound with the haemoglobin is proved by the fact that haemoglobin thoroughly saturated with oxygen (oxyhaemoglobin) has a definite absorption spectrum showing two bands between the D and E lines, whilst haemoglobin from which the oxygen has been completely removed only gives one band between those lines.
The actual state of acid-base balance, then, can only be determined by the use of any two of a number of interdependent variables, such as total CO_{2}, CO_{2} tension, pH, H_{2}CO_{3} concentration, other buffers than bicarbonate, plasma chloride, ratio of oxyhaemoglobin to haemoglobin, etc.
